|Figure 5: Monitorization of the effect of different
agonists on [Ca2+]c in pancreatic acinar cells
cultured for 48 hours. Pancreatic acinar cells were allowed to
grow for 48 hours. Then cells were loaded with fura-2 and
fluorescence was analyzed after stimulation of cells with 1 nM
CCK-8 (A), 100 nM ACh (B) or 20 pM CCK-8 (C). The changes in
[Ca2+]c did not follow the typical response pattern
shown by pancreatic acinar cells at day zero of culture. Viability
of cells was not decreased under culture conditions and in the
absence of stimulus. Stimuli were added to the cells on the day in
which the tests for Ca2+ mobilization were performed.
Traces are representative of 22–35 cells analyzed in 5-6
independent experiments for each treatment.
