Relapsedandrefractorydiseaseshortensthesurvivalofacutemyeloidleukemia(AML)patients.Therefore,additional therapyapartfromconventionalchemotherapyandstemcelltransplantationisurgentlyneeded,especiallyineradicatingresidual leukemic stem cells. The identification of leukemia associated antigens, and the observation that administration of allogeneic T cellsmaymediategraftversusleukemiaeffectpavedthewaytodevelopvariousimmunotherapystrategies.Effectiveprovisionof bothtumorantigenandco-stimulationareessentialforstrategiesaimedatenrollingcytotoxicT-lymphocytestoeradicateleukemic cells.Inthisstudy,weattemptedtoup-regulateco-stimulatorsonleukemiccells(termed“dendritisation”tomimicdendriticcells inprovidingstrongco-stimulation)andcouplewithinherentleukemicantigenstogeneratespecificantileukemicT-lymphocytes againstautologousblastcells.OurstudyshowsevidenceofdistinctcytotoxicityofCD4positiveTlymphocytesagainstautologous blastcells.DendritisedAMLblastswereabletofunctionwithantigenpresentingcapability,whilestillphenotypicallyexistingas blastcells.ThiswasalsoachievableforawiderangeofsubtypesofAML,makingitapotentialimmunotherapyagainstAML
